扁桃斑鸠菊抗乳腺癌活性部位筛选研究Screening of anti-breast cancer active fractions of Vernonia amygdalina

摘要(Abstract):
本研究为了筛选出扁桃斑鸠菊的抗乳腺活性部位,以人乳腺癌细胞株(MCF-7)为实验模型,采用CCK-8实验评价扁桃斑鸠菊提取物对乳腺癌细胞增殖的抑制作用。选择极性不同的溶剂,设计了三种不同的提取路线,最终得到极性不同的各个部位提取物(VA1-VA8)。大量提取活性较好的乙酸乙酯提取物,然后洗脱分离为6个段(A2-A7),继续运用CCK-8细胞毒性实验来追踪抗乳腺癌活性最佳的部位。CCK-8实验结果显示:扁桃斑鸠菊乙酸乙酯提取物对MCF-7细胞增殖表现出良好的抑制作用,且呈剂量依赖性。在A2-A7的6个洗脱产物中,浓度为100μg/mL时,50%乙酸乙酯-石油醚洗脱段(A5)对人乳腺癌细胞MCF-7具有最佳的抑制效果,抑制率为83.34%。
